Aging results partly from development of patterns of gene expression that determine specific complexes of proteins and lipids that become progressively less adequate for maintenance of brain. Aging also results from progressive uncontrolled oxidation, causing DNA damage, mitochondrial decay, chronic inflammation, and excessive cell death in brain. Data from cell, mouse, rat, and monkey models, indicate that caloric restriction (CR) inhibits development of age-related proteins and lipids, and inhibits oxidative damage by affecting expression of multiple genes that are tightly conserved across mammals, including PPARs, SREBPs, NFkB, 17BHSD, HO-1, GCSh, TNFax, and VDAC-1 and results in inhibition of cognitive decline, inhibition of multiple diseases and increased lifespan. Several nutrients, including docosahexanoic acid, resveratrol, n-acetyl cysteine, R+ lipoic acid, and acetyl-L carnitine mimic some CR effects on gene expression. These findings offer unique support to the more macroscopic and more controversial data from medical, biochemical, nutrition, and aging research that the same nutrients can inhibit a wide spectrum of disorders, including declines of cognition, learning, memory, motor function, balance and reaction speed, and offer firm support for their use to combat brain aging. |
